Although she functioned as a Queen, her extreme age and size placed her closer to an Empress-like figure in the hive hierarchy. Dr. Groves described her as ancient, worn by time, yet still alive and aware after an almost unimaginable period of captivity. Thousands of years before the events of the game, the Matriarch was captured by the Yautja and imprisoned on BG-386. The Predators used her as a source of Xenomorph eggs, allowing them to breed new Aliens for ritual hunts. Even after the Yautja city fell into ruin, the Matriarch remained alive inside the ancient complex. The Matriarch escaped after Specimen 6 broke free from containment and sabotaged Weyland-Yutani's systems. Specimen 6 released other Xenomorphs inside the complex and eventually freed the Queen from her restraints. Once loose, the Matriarch and her brood fled the research facility and moved toward the nearby Freya's Prospect colony. The escape transformed the outbreak from a contained laboratory disaster into a full planetary crisis. With the Queen free, the Xenomorphs could expand rapidly, harvest hosts, and establish a functioning hive outside Weyland-Yutani control.

The Matriarch survived several Marine assaults, including a major attack led by Major Van Zandt. During the Alien campaign, she also directed Specimen 6 through the hive mind, ordering her to kill, harvest, sabotage communications, and fight both Marines and Predators on BG-386.

The Matriarch was far older and grayer than most Queens, likely because of her extreme age. Dr. Groves described her as an intelligence that had remained captive and aware for tens of thousands of years. Her crest and face carried visible battle scars, giving her a worn and ancient appearance compared to younger Queens. She also possessed an exceptionally tough hide that could resist conventional gunfire. During the Marine battle against her, direct ballistic attacks were ineffective, forcing Rookie to rely on the refinery's explosive systems instead. Like other Queens, she had immense strength, a massive tail, large claws, and the ability to produce eggs through her ovipositor.

The Matriarch was finally confronted by Rookie, the Colonial Marine protagonist, inside her refinery chamber. With help from Corporal Tequila over the radio, Rookie used the environment against the Queen rather than trying to kill her with ordinary weapons. He detonated combustible containers around the chamber, gradually weakening her and triggering a large explosion. The blast incinerated the Matriarch and ended her reign over the hive on BG-386. Her death caused a neural shock throughout her brood, temporarily paralyzing many of the surrounding Xenomorph Warriors with pain.

The Specimen 6 Queen from the end of Aliens vs. Predator 2010

The Matriarch shared similarities with several other unusually powerful Xenomorph Queens from Alien and Predator lore. One of the closest examples was the Antarctica Queen from Alien vs. Predator (2004), another Queen imprisoned by the Yautja and used to produce eggs for ritual hunts.
